About

role
Activating kinases are frequently present in tumor that originate or metastasize to the brain. In this project we will optimally adapt therapies to multiple activated kinases. You will work on the mechanisms that underly effective anti-cancer kinase inhibitors and delineate them by performing drug screens aimed at individual targets followed by in vivo validation of the results. Pharmacodynamic, pharmacokinetic and blood brain barrier studies of existing and de novo generated drugs will be performed in collaboration with two companies. You will also work on reporter systems for real time assessment therapy resistance.

The
project
Activating kinases are frequently present in tumor that originate or metastasize to the brain. In this project we will optimally adapt therapies to multiple activated kinases. For this, we have generated a target predictor for each small molecule inhibitor based on the drug and target structures. This enables us to match bioactivity (target) fingerprints to personalized vulnerabilities aimed to generate more effective therapies. Within the project, we aim to test existing but also de novo generated kinase inhibitors for their effect against brain tumors. Targeting multiple targets comes with the cost of potential toxic effects. In addition to the target predictor, we have generated a predictor for selecting optimal target-combinations with limited adverse events. Our approach will guide us to reach an optimal effectivity to toxicity ratio for existing and new small molecules.

About
you
You are a highly motivated and ambitious candidate with a HLO (hoger laboratorium onderwijs) education or a bachelor/master degree in biomedical sciences or related field. Preferably you have a Article 9 or 13 degree to allow for animal experimentation or have the wish to obtain it within 3 months of the start of the project.

In addition you meet the following requirements:
  • You like to work with animals since animal experiments are estimated to be 40% of the work-load.
  • You are a well-organized person that is not afraid of working within technically challenging environment; you are experienced in primary cell culture and in animal studies and their registration;
  • You have strong analytical attitude and the ability to express yourself firmly and adequately;
  • You have affinity with modern molecular biology and next-generation techniques (CRISPR technology, next-generation sequencing, mass cytometry);
  • You have experience with reporting through presenting data and figures, writing reports and protocols; you are able to function well in a team as well as independently within an international cross-disciplinary environment;
  • You are proficient in English, both verbal as in writing. For international applicants, an English proficiency test score or a recommendation by a referee is desired.
Our
offer
Depending on your experience, we offer a competitive salary of a minimum of € 2,367 to a maximum of € 3,606 for a full working week of 36 hours.
  • Additional benefits include an 8.3% end-of-year bonus and 8.0% holiday pay.
  • The opportunity to follow further training and development.
  • Pension is accrued at the ABP. VUmc contributes a large part of the premium (70%).
  • In addition to excellent accessibility by public transport, we reimburse 75% of your OV travel costs (commuting costs).
We offer a contract for the duration of 4 years with a mid-term evaluation after 12 months.
